In vitro | NEP, evaluated at concentrations of 1-30 μM over a period of 6 hours, exhibits minimal to no cytotoxic effects on HepG2 and PC12 cells[1]. When PC12 cells, pre-treated with varying concentrations of 6-OHDA (0, 50, 100, and 200 μM for 30 minutes), are exposed to 10 μM of NEP for 4 hours, a progressive decline in fluorescence intensity is observed[1]. The chemical structure of NEP includes a dithiarsolane moiety (a five-membered As-S ring), which serves as the binding site for VDPs. Upon interaction with VDPs, NEP generates a robust green fluorescence signal, resulting from the cleavage of the cyclic dithiarsolane and a subsequent intramolecular cyclization that releases the fluorophore. This fluorescence response of NEP remains stable across a pH range of 7 to 10[1]. |
