Verified Activity
1. 25 μg total protein per lane of various lysates (see on figure) probed with RPE65 monoclonal antibody, unconjugated (TMAB-12352) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r. T. for 60 min.
2. Paraformaldehyde-fixed, paraffin embedded Rat Eye; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with RPE65 Monoclonal Antibody, Unconjugated (TMAB-12352) at 1: 200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L Secondary Antibody-HRP and DAB staining.

Plays important roles in the production of 11-cis retinal and in visual pigment regeneration. The soluble form binds vitamin A (all-trans-retinol), making it available for LRAT processing to all-trans-retinyl ester. The membrane form, palmitoylated by LRAT, binds all-trans-retinyl esters, making them available for IMH (isomerohydrolase) processing to all-cis-retinol. The soluble form is regenerated by transferring its palmitoyl groups onto 11-cis-retinol, a reaction catalyzed by LRAT. The enzymatic activity is linearly dependent of the expression levels and membrane association.
