Verified Activity
1. Tissue/cell: rat pancreas tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer (0.01 M, pH 6.0), Boiling bathing for 15 min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30 min; Blocking buffer (normal goat serum) at 37℃ for 20 min;
Incubation: Anti-Insulin Polyclonal Antibody, Unconjugated (TMAB-11776) 1: 200, overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining
2. Tissue/cell: rat pancreas tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer (0.01 M, pH 6.0), Boiling bathing for 15 min; Blocking buffer (normal goat serum) at 37℃ for 20 min;
Incubation: Anti-Insulin Polyclonal Antibody, Unconjugated (TMAB-11776) 1: 200, overnight at 4°C; The secondary antibody was Goat Anti-Rabbit IgG, FITC conjugated used at 1: 200 dilution for 40 minutes at 37°C.
3. Mouse Pancreas (C57BL/6 strain) fixed in 4% PFA for 2 hours at RT, then transferred to 30% sucrose overnight at 4°C. Treated with TBS containing 5% goat serum and 0.3% Triton X-100, to block unspecific binding sites and permeabilize cell membranes, for 30 min at RT. Antibody (TMAB-11776-FITC) diluted at 1:100 in TBS containing 2% goat serum applied and incubated overnight at 4°C. Rinse sections in TBS 2x5min on a shaker, mount section with vectorshield (with DAPI), then cover them with coverslips and seal the edge of the coverslip on the slide with nail polish.

Paraformaldehyde-fixed, paraffin embedded (rat pancreas tissue); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30 min; Antibody incubation with (Proinsulin) Polyclonal Antibody, Unconjugated (TMAB-11776) at 1:400 overnight at 4°C, followed by a conjugated secondary for 20 minutes and DAB staining.
5. Paraformaldehyde-fixed, paraffin embedded (rat pancreas); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30 min; Antibody incubation with (Proinsulin) Polyclonal Antibody, Unconjugated (TMAB-11776) at 1: 200 overnight at 4°C, followed by operating according to SP Kit (Rabbit) instructions and DAB staining.
