Paraformaldehyde-fixed, paraffin embedded (Human stomach cancer); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30 min; Antibody incubation with (phospho-LYRIC (Ser568)) Polyclonal Antibody, Unconjugated (TMAB-10925) at 1:400 overnight at 4°C, followed by operating according to SP Kit (Rabbit) instructions and DAB staining.

Research BackgroundLYRIC is a type II membrane protein, the precise role of which has yet to be defined. It is expressed in breast and prostate carcinoma and is a potential mediator of lung metastasis. It has also been found in mouse Purkinje neurons and cerebellum. In epithelial cells it is recruited to tight junctions (TJ) during the maturation of the TJ complexes. LYRIC co-localizes with beta-catenin in normal and tumour tissues.

Function
Downregulates SLC1A2/EAAT2 promoter activity when expressed ectopically. Activates the nuclear factor kappa-B (NF-kappa-B) transcription factor. Promotes anchorage-independent growth of immortalized melanocytes and astrocytes which is a key component in tumor cell expansion. Promotes lung metastasis and also has an effect on bone and brain metastasis, possibly by enhancing the seeding of tumor cells to the target organ endothelium. Induces chemoresistance.
