| Verified Activity | 1. Sample: Testis (Mouse) Lysate at 40 μg Primary: Anti-PBK (TMAB-10025) at 1/1000 dilution Secondary: IRDye800CW Goat Anti-Rabbit IgG at 1/20000 dilution Predicted band size: 36 kD Observed band size: 37 kD 2. Blank control (black line): HepG2. Primary Antibody (green line): Rabbit Anti-PBK antibody (TMAB-10025) Dilution: 2 μg/Test; Secondary Antibody (white blue line): Goat anti-rabbit IgG-FITC Dilution: 0.5 μg/Test. Isotype control (orange line): Normal Rabbit IgG Protocol The cells were fixed with 4% PFA (10 min at room temperature) and then permeabilized with 90% ice-cold methanol for 20 min at -20℃, The cells were then incubated in 5% BSA to block non-specific protein-protein interactions for 30 min at room temperature. Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed. |

| Research Background | This gene encodes a serine/threonine protein kinase related to the dual specific mitogen-activated protein kinase kinase (MAPKK) family. Evidence suggests that mitotic phosphorylation is required for its catalytic activity. The encoded protein may be involved in the activation of lymphoid cells and support testicular functions, with a suggested role in the process of spermatogenesis. Overexpression of this gene has been implicated in tumorigenesis. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2013] |
| Immunogen | KLH conjugated synthetic peptide: human PBK |
| Antigen Species | Human |
| Gene Name | PBK |
| Gene ID | |
| Protein Name | Lymphokine-activated killer T-cell-originated protein kinase |
| Uniprot ID | |
| Function | Phosphorylates MAP kinase p38. Seems to be active only in mitosis. May also play a role in the activation of lymphoid cells. When phosphorylated, forms a complex with TP53, leading to TP53 destabilization and attenuation of G2/M checkpoint during doxorubicin-induced DNA damage. |
