11. 25 μg total protein per lane of various lysates (see on figure) probed with PAX8 monoclonal antibody, unconjugated (TMAB-10020) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r. T. for 60 min.
12. 4% Paraformaldehyde-fixed OVCAR-3 (H) cell; Triton X-100 at r. T. for 20 min; Antibody incubation with (PAX8) monoclonal Antibody, unconjugated (TMAB-10020) 1:100, 90 min at 37°C; followed by conjugated Goat Anti-Rabbit IgG antibody (green) at 37°C for 90 min, DAPI (blue) was used to stain the cell nuclei. PBS instead of the primary antibody was used as the blank control.
13. The OVCAR-3 (H) cells were fixed with 4% PFA (10 min at r. T.) and then permeabilized with 90% ice-cold methanol for 20 min at -20℃, the cells then were incubated in 5% BSA to block non-specific protein-protein interactions (30 min at r. T.). Primary Antibody (green): Rabbit Anti-PAX8 antibody (TMAB-10020, 1:100); Secondary Antibody (white blue): Goat anti-Rabbit IgG-BF488: 1 μg/test. Isotype Control (orange): Rabbit IgG. Blank control (black): PBS. Acquisition of 20,000 events was performed.

Research BackgroundThis protein is a member of the paired box (PAX) family of transcription factors, typically containing a paired box domain, an octapeptide, and a paired-type homeodomain. This family plays critical roles during fetal development and cancer growth. The specific function of the PAX8 is unknown but it may involve kidney cell differentiation, thyroid development, or thyroid dysgenesis. Alternative splicing in the gene by inclusion or exclusion of exons 7 and/or 8 has produced several known products but the biological significance of the variants is unknown. Several other splice variants have been proposed but the full nature of these products has not been described. Pax8 is also a marker of otic progenitor cells.
