Verified Activity | 1. Immunochemical staining of human HSP12A in human colon carcinoma with rabbit monoclonal antibody (1:200, formalin-fixed paraffin embedded sections). Positive staining was localized to colonic gland. 2. Immunochemical staining of human HSP12A in human cirrhosis with rabbit monoclonal antibody (1:200, formalin-fixed paraffin embedded sections). Positive staining was localized to hepatocyte. 3. Anti-HSP12A rabbit monoclonal antibody at 1:500 dilution. -Lane A: 293T Whole Cell Lysate. -Lane B: Hela Whole Cell lysate. -Lysates/proteins at 30 μg per lane. -Secondary -Goat Anti-Rabbit IgG H&L (Dylight800) at 1/10000 dilution. -Developed using the Odyssey technique. -Performed under reducing conditions. -Predicted band size:111 kDa. -Observed band size:170 kDa. 4. HSP12A was immunoprecipitated using: -Lane A:0.5 mg MCF-7 Whole Cell Lysate. -Lane B:0.5 mg 293T Whole Cell Lysate. -Lane C:0.5 mg Hela Whole Cell Lysate. -2 µL anti-HSP12A rabbit monoclonal antibody and 15 μl of 50 % Protein G agarose. -Primary antibody: -Anti-HSP12A rabbit monoclonal antibody, at 1:100 dilution. -Secondary antibody: -Dylight 800-labeled antibody to rabbit IgG (H+L), at 1:5000 dilution. -Developed using the odyssey technique. -Performed under reducing conditions. -Predicted band size: 111 kDa. -Observed band size: 111 kDa. 5. Immunofluorescence staining of ORP150 in MCF7 cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti-Human ORP150 monoclonal antibody (dilution ratio 1:60) at 4°C overnight. Then cells were stained with the Alexa Fluor®488-conjugated Goat Anti-rabbit IgG secondary antibody (green). Positive staining was localized to Cytoplasm. |

Research Background | Hypoxia up-regulated protein 1, also known as 15 kDa oxygen-regulated protein, 17 kDa glucose-regulated protein, ORP-15, GRP-17, and HYOU1, is a member of the heat shock protein 7 family. Seven members from four different heat shock protein (HSP) families were identified including HYOU1 (ORP15), HSPC1 (HSP86), HSPA5 (Bip), HSPD1 (HSP6), and several isoforms of the two testis-specific HSP7 chaperones HSPA2 and HSPA1L. HYOU1 is highly expressed in tissues that contain well-developed endoplasmic reticulum and synthesize large amounts of secretory proteins. It is highly expressed in the liver and pancreas. HYOU1 is also expressed in macrophages within aortic atherosclerotic plaques and in breast cancers. HYOU1 has a pivotal role in cytoprotective cellular mechanisms triggered by oxygen deprivation. It may play a role as a molecular chaperone and participate in protein folding. Suppression of HYOU1 is associated with accelerated apoptosis. It is suggested to have an important cytoprotective role in hypoxia-induced cellular perturbation. This protein is up-regulated in tumors, especially in breast tumors, and thus it is associated with tumor invasiveness. |
