Human embryonic stem (ES) cells were fixed and permeabilized with BD Cytofix/Cytoperm™ Fixation/Permeabilization buffer (BD Pharmingen™ Cat. No. 554714), and stained with Purified Mouse anti-Oct4 monoclonal antibody, then a FITC-conjugated second step antibody. The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.

POU5F1 is a transcription factor and master regulator of cell pluripotency with indispensable roles in early embryo development and cell lineage specification. The role of embryonic POU5F1 in blastocyst formation and cell lineage specification differs between mammalian species.
