1. Sample:
Raji (Human) Cell Lysate at 30 μg
Primary: Anti-NRSF (TMAB-01273) at 1/300 dilution
Secondary: IRDye800CW Goat Anti-Rabbit IgG at 1/20000 dilution
Predicted band size: 121 kDa
Observed band size: 121 kDa
2. Tissue/cell: human gastric tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer (0.01M, pH6.0), Boiling bathing for 15 min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30 min; Blocking buffer (normal goat serum) at 37°C for 20 min;
Incubation: Anti-NRSF/REST Polyclonal Antibody, Unconjugated (TMAB-01273) 1:200, overnight at 4°C, followed by conjugation to the secondary antibody and DAb staining.
3. NRSF was immunoprecipitated from human hela cells lysate with TMAB-01273 at 1/150 dilution. Western blot was performed from the immunoprecipitate using protein A/G beads. HRP Conjugated Mouse anti-Rabbit IgG (Light Chain specific) was used as secondary antibody at 1:5000 dilution.

Research BackgroundThis gene encodes a transcriptional repressor that represses neuronal genes in non-neuronal tissues. It is a member of the Kruppel-type zinc finger transcription factor family. It represses transcription by binding a DNA sequence element called the neuron-restrictive silencer element. The protein is also found in undifferentiated neuronal progenitor cells and it is thought that this repressor may act as a master negative regular of neurogenesis. Alternatively spliced transcript variants have been described [provided by RefSeq, Jul 2010]

Function
Transcriptional repressor which binds neuron-restrictive silencer element (NRSE) and represses neuronal gene transcription in non-neuronal cells. Restricts the expression of neuronal genes by associating with two distinct corepressors, mSin3 and CoREST, which in turn recruit histone deacetylase to the promoters of REST-regulated genes. Mediates repression by recruiting the BHC complex at RE1/NRSE sites which acts by deacetylating and demethylating specific sites on histones, thereby acting as a chromatin modifier. Transcriptional repression by REST-CDYL via the recruitment of histone methyltransferase EHMT2 may be important in transformation suppression.
