Verified Activity | 1. Immunofluorescence staining of hela cell with TMAH-00758 at 1:60, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was FITC-conjugated AffiniPure Goat Anti-Human IgG(H+L). 2. Immunofluorescence staining of U251 cell with TMAH-00758 at 1:60, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was FITC-conjugated AffiniPure Goat Anti-Human IgG(H+L). 3. Overlay Peak curve showing PC3 cells stained with TMAH-00758 (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*10^6 cells) for 45min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-human IgG(H+L) at 1:200 dilution for 35min at 4°C.Control antibody (green line) was human IgG (1ug/1*10^6 cells) used under the same conditions. Acquisition of >10,000 events was performed. |

Research Background | Pro-inflammatory cytokine. Involved in the innate immune response to bacterial pathogens. The expression of MIF at sites of inflammation suggests a role as mediator in regulating the function of macrophages in host defense. Counteracts the anti-inflammatory activity of glucocorticoids. Has phenylpyruvate tautomerase and dopachrome tautomerase activity (in vitro), but the physiological substrate is not known. It is not clear whether the tautomerase activity has any physiological relevance, and whether it is important for cytokine activity. |
