1. Paraformaldehyde-fixed, paraffin embedded (human stomach); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30 min; Incubation with (MCM7) Monoclonal Antibody, Unconjugated (TMAB-08673) at 1:100 overnight at 4°C, followed by operating according to SP Kit (Rabbit) instructions and DAB staining.
2. Paraformaldehyde-fixed, paraffin embedded Human lung squamous cell carcinoma; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Antibody incubation with MCM7 Monoclonal Antibody, Unconjugated (TMAB-08673) at 1:150 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L Secondary Antibody-HRP and DAB staining.
3. 5. Paraformaldehyde-fixed, paraffin embedded Human Fundus; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Antibody incubation with MCM7 Monoclonal Antibody, Unconjugated (TMAB-08673) at 1:150 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L Secondary Antibody-HRP and DAB staining.
6. 25 μg total protein per lane of various lysates (see on figure) probed with MCM7 monoclonal antibody, unconjugated (TMAB-08673) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r. T. for 60 min.

DNA replication licensing factor MCM7

Acts as component of the MCM2-7 complex (MCM complex) which is the putative replicative helicase essential for 'once per cell cycle' DNA replication initiation and elongation in eukaryotic cells. The active ATPase sites in the MCM2-7 ring are formed through the interaction surfaces of two neighboring subunits such that a critical structure of a conserved arginine finger motif is provided in trans relative to the ATP-binding site of the Walker A box of the adjacent subunit. The six ATPase active sites, however, are likely to contribute differentially to the complex helicase activity. Required for S-phase checkpoint activation upon UV-induced damage.
