| Verified Activity | 1. 4% Paraformaldehyde-fixed MCF-7 (H) cell; Triton X-100 at r. T. for 20 min; Antibody incubation with (MCM4) monoclonal Antibody, unconjugated (TMAB-08669) 1:100, 90 min at 37°C; followed by conjugated Goat Anti-Rabbit IgG antibody (green) at 37°C for 90 min, DAPI (blue) was used to stain the cell nuclei. PBS instead of the primary antibody was used as the blank control. 2. 25 μg total protein per lane of various lysates (see on figure) probed with MCM4 monoclonal antibody, unconjugated (TMAB-08669) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r. T. for 60 min. |

| Function | Acts as component of the MCM2-7 complex (MCM complex) which is the putative replicative helicase essential for 'once per cell cycle' DNA replication initiation and elongation in eukaryotic cells. The active ATPase sites in the MCM2-7 ring are formed through the interaction surfaces of two neighboring subunits such that a critical structure of a conserved arginine finger motif is provided in trans relative to the ATP-binding site of the Walker A box of the adjacent subunit. The six ATPase active sites, however, are likely to contribute differentially to the complex helicase activity. |
