| Verified Activity | 1. Western blot analysis of A431 cell lysate. Using MAD1 (TMAB-08496) monoclonal antibody at 1: 1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r. T. for 60 min. 2. The Hela (H) cells were fixed with 4% PFA (10 min at r. T.) and then permeabilized with 90% ice-cold methanol for 20 min at -20℃, the cells then were incubated in 5% BSA to block non-specific protein-protein interactions (30 min at r. T.), followed by secondary antibody incubation for 40 min at room temperature. Primary Antibody (green): Rabbit Anti-MAD1 antibody (TMAB-08496, 1: 100); Isotype Control (orange): Rabbit IgG. Blank control (black): PBS. Acquisition of 20,000 events was performed. |

| Subcellular Localization | Nucleus. Chromosome, centromere, kinetochore. Cytoplasm, cytoskeleton, centrosome. Cytoplasm, cytoskeleton, spindle. Note=From the beginning to the end of mitosis, it is seen to move from a diffusely nuclear distribution to the centrosome, to the spindle midzone and finally to the midbody. Co-localizes with NEK2 at the kinetochore. |
| Tissue Specificity | Expressed weakly at G0/G1 and highly at late S and G2/M phase. |
| Construction | Recombinant Antibody |
| Purification | Protein A purified |
| Appearance | Liquid |
| Formulation | 10mM phosphate buffered saline (pH 7.4) with 150mM sodium chloride, 0.05% BSA, 0.02% Proclin300 and 50% glycerol. |
| Research Background | Component of the spindle-assembly checkpoint that prevents the onset of anaphase until all chromosomes are properly aligned at the metaphase plate.Forms a heterotetrameric complex with the closed conformation form of MAD2L1 (C-MAD2) at unattached kinetochores during prometaphase, recruits an open conformation of MAD2L1. |
| Immunogen | A synthesized peptide: human MAD1 |
| Antigen Species | Human |
| Gene Name | MAD1L1 |
| Gene ID | |
| Protein Name | Mitotic spindle assembly checkpoint protein MAD1 |
| Uniprot ID | |
| Function | Component of the spindle-assembly checkpoint that prevents the onset of anaphase until all chromosomes are properly aligned at the metaphase plate. May recruit MAD2L1 to unattached kinetochores. Has a role in the correct positioning of the septum. Required for anchoring MAD2L1 to the nuclear periphery. Binds to the TERT promoter and represses telomerase expression, possibly by interfering with MYC binding. |
| Molecular Weight | Theoretical: 83 kDa. Actual: 80 kDa. |
