| Verified Activity | 1. Blank control: HepG2. Primary Antibody (green line): Rabbit Anti-Livin antibody (TMAB-08332) Dilution: 1 μg /10^6 cells; Isotype Control Antibody (orange line): Rabbit IgG. Secondary Antibody: Goat anti-rabbit IgG-AF647 Dilution: 1 μg /test. Protocol The cells were fixed with 4% PFA (10 min at room temperature) and then permeabilized with 0.1% PBST for 20 min at room temperature. The cells were then incubated in 5% BSA to block non-specific protein-protein interactions for 30 min at room temperature. Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed. 2. Blank control (blue line): Hep G2 (fixed with 70% ethanol (Overmight at 4℃) and then permeabilized with 90% ice-cold methanol for 30 min at -20℃). Primary Antibody (green line): Rabbit Anti-Nanog antibody (TMAB-08332), Dilution: 1 μg /10^6 cells; Isotype Control Antibody (orange line): Rabbit IgG. Secondary Antibody (white blue line): Goat anti-rabbit IgG-PE, Dilution: 1 μg /test. 3. Paraformaldehyde-fixed, paraffin embedded (Rat skin); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30 min; Antibody incubation with (Livin) Polyclonal Antibody, Unconjugated (TMAB-08332) at 1:400 overnight at 4°C, followed by operating according to SP Kit (Rabbit) instructions and DAB staining. 4. | Research Background | Livin is a novel member of the IAP protein family and contains a single baculoviral IAP repeat (BIR) domain and a RING finger domain and has two isoforms termed Livin alpha and Livin beta. The BIR domain is essential for inhibitory activity and interacts with caspases, while the RING finger domain sometimes enhances antiapoptotic activity but does not inhibit apoptosis alone. The two isoforms of Livin appear to have different functions and tissue distributions. |

| Function | Protects against apoptosis induced by TNF or by chemical agents such as adriamycin, etoposide or staurosporine. Suppression of apoptosis is mediated by activation of MAPK8/JNK1, and possibly also of MAPK9/JNK2. This activation depends on TAB1 and NR2C2/TAK1.
