Verified Activity | 1. Immunofluorescence staining of Human LTA4H in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS, blocked with 10% serum, and incubated with rabbit anti-Human LTA4H monoclonal antibody (1:300) at 4°C overnight. Then cells were stained with the Alexa Fluor® 488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI (blue). Positive staining was localized to cytoplasm. 2. Anti-LTA4H rabbit monoclonal antibody at 1:500 dilution. -Lane A: Hela Whole Cell Lysate. -Lane B: A549 Whole Cell Lysate. -Lane C: 293T Whole Cell lysate. -Lysates/proteins at 30 μg per lane. -Secondary -Goat Anti-Rabbit IgG H&L (Dylight800) at 1/10000 dilution. -Developed using the Odyssey technique. -Performed under reducing conditions. -Predicted band size:69 kDa. -Observed band size:69 kDa. 3. LTA4H was immunoprecipitated using: -Lane A:0.5 mg Hela Whole Cell Lysate. -Lane B:0.5 mg 293T Whole Cell Lysate -0.5 µL anti-LTA4H rabbit monoclonal antibody and 15 μl of 50 % Protein G agarose. -Primary antibody: -Anti-LTA4H rabbit monoclonal antibody, at 1:500 dilution. -Secondary antibody: -Dylight 800-labeled antibody to rabbit IgG (H+L), at 1:5000 dilution. -Developed using the odyssey technique. -Performed under reducing conditions. -Predicted band size: 110 kDa. -Observed band size: 110 kDa |

Research Background | Leukotriene A-4 hydrolase, also known as LTA-4 hydrolase, Leukotriene A (4) hydrolase, LTA4H, and LTA4, is a cytoplasm protein that belongs to the peptidase M1 family. LTA4H hydrolyzes an epoxide moiety of leukotriene A4 (LTA-4) to form leukotriene B4 (LTB-4). This enzyme also has some peptidase activity. The leukotrienes (LTs) are a class of structurally related lipid mediators involved in the development and maintenance of inflammatory and allergic reactions. In the biosynthesis of LTs, arachidonic acid was converted into the unstable intermediate epoxide LTA4, which may, in turn, be conjugated with glutathione to form the spasmogenic LTC4, or hydrolyzed into the pro-inflammatory lipid mediator LTB4 in a reaction catalyzed by Leukotriene A4 hydrolase (LTA4H). LTB4 is a classical chemoattractant of human neutrophils and triggers adherence and aggregation of leukocytes to vascular endothelium, and also modulates immune responses. As a bifunctional zinc metalloenzyme, LTA4H also exhibits an anion-dependant arginyl aminopeptidase activity of high efficiency and specificity in addition to its epoxide hydrolase activity. LTA4H is regarded as a therapeutic target for inflammation. |
