1. Paraformaldehyde-fixed, paraffin embedded (human tonsil); Antigen retrieval by boiling in sodium EDTA buffer (Ph9.0) for 15 min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30 min; Incubation with (IL18) Monoclonal Antibody, Unconjugated (TMAB-07619) at 1: 250 overnight at 4°C, followed by operating according to SP Kit (Rabbit) instructions and DAB staining.
2. Sample:
Lane 1: Normal HeLa cell lysates
Lane 2: HeLa cell treated with Brefeldin A (5 μg/ml, 15 min)
Lane 3: Jurkat cell lysates (Negative control)
Primary: Anti-IL18 (TMAB-07619) at 1/1000 dilution
Secondary: IRDye800CW Goat Anti-Rabbit IgG at 1/20000 dilution

3. 4. The Hela (H) cells were fixed with 4% PFA (10 min at r. T.) and then permeabilized with 90% ice-cold methanol for 20 min at -20℃, the cells then were incubated in 5% BSA to block non-specific protein-protein interactions (30 min at r. T.), followed by secondary antibody incubation for 40 min at room temperature. Primary Antibody (green): Rabbit Anti-IL18 antibody (TMAB-07619, 1:100); Isotype Control (orange): Rabbit IgG. Blank control (black): PBS. Acquisition of 20,000 events was performed.
5. Paraformaldehyde-fixed, paraffin embedded Human Chronic tonsillitis; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with IL18 Monoclonal Antibody, Unconjugated (TMAB-07619) at 1: 200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L Secondary Antibody-HRP and DAB staining.
6. Paraformaldehyde-fixed, paraffin embedded Human Pyogenic appendicitis; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with IL18 Monoclonal Antibody, Unconjugated (TMAB-07619) at 1: 200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L Secondary Antibody-HRP and DAB staining.
