| Verified Activity | 1. Paraformaldehyde-fixed, paraffin embedded Human Tonsil; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with Human IgM Monoclonal Antibody, Unconjugated (TMAB-07445) at 1:200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L-HRP and DAB staining. 2. Paraformaldehyde-fixed, paraffin embedded Human Lymph; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with Human IgM Monoclonal Antibody, Unconjugated (TMAB-07445) at 1:200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L-HRP and DAB staining. 3. Paraformaldehyde-fixed, paraffin embedded Human Spleen; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with Human IgM Monoclonal Antibody, Unconjugated (TMAB-07445) at 1:200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L-HRP and DAB staining. 4. 25 ug total protein per lane of various lysates (see on figure) probed with Human IgM monoclonal antibody, unconjugated (TMAB-07445) at 1: 1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r. T. for 60 min. |

| Research Background | IgM normally constitutes about 10% of serum immunoglobulins. IgM antibody is prominent in early immune responses to most antigens and is largely confined to plasma due to it's large size. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. Due to it's high valency IgM is more efficient than other isotypes is binding antigens with repeating epitopes (virus particles and red blood cells) and is more efficient than IgG in activiating the complement pathway. The gene for the mu constant region contains four domains separated by short intervening sequences. |
