| Verified Activity | 1. Comparative His-tag detection by Western blot was performed using Anti-His tag Antibody (3T670) (TMAS-00127) and Mouse Anti-His mAb (Competitor B). Both antibodies were used to probe the same cell lysates containing His-tagged fusion protein. The signal was developed with Goat Anti-Mouse IgG (H&L) [HRP] Polyclonal Antibody. 2. Immunoprecipitates from cell lysates containing His-tagged fusion protein were analyzed by Western blot using Anti-His tag Antibody (3T670) (TMAS-00127). Lane 1. Positive control containing His-tagged fusion protein. Lane 2. Negative control – IP with isotype control antibody. Lane 3. Immunoprecipitation with Anti-His tag Antibody (3T670) (TMAS-00127). 3. His-tag was detected in Multiple Tag Cell Lysate by Western blot using Anti-His tag Antibody (3T670) (TMAS-00127). The signal was developed with Goat Anti-Mouse IgG (H&L) [HRP] Polyclonal Antibody. His-tagged fusion protein: Predicted MW: 52 kDa Observed MW: 52 kDa 4. Comparative His-tag detection by Dot blot was performed using Anti-His tag Antibody (3T670) (TMAS-00127) and two Mouse Anti-His mAbs (Competitor 1, Competitor 2). All three antibodies were used to probe the same samples containing His-tagged fusion protein. The signal was developed with Goat Anti-Mouse IgG (H&L) [HRP] Polyclonal Antibody. 5. Lot-to-lot consistency of antibody performance was analyzed for 4 batches of Anti-His tag Antibody (3T670) (TMAS-00127) by Western blot. Detection of His-tag in CHO cells transfected with His-tagged protein (Green), and non-transfected CHO cells (Black) by flow cytometry using Anti-His tag Antibody (3T670) (TMAS-00127). The signal was developed with FITC conjugated Goat Anti-Mouse IgG. 7. Comparative His-tag detection by Western blot was performed using Anti-His tag Antibody (3T670) (TMAS-00127) and Mouse Anti-His mAb (Competitor A). Both antibodies were used to probe the same sample containing overexpressed His-tagged fusion protein. 8. Detection of N- or C-terminal His tags in recombinant fusion proteins were analyzed by Western blot using Anti-His tag Antibody (3T670) (TMAS-00127). Lane 1: N-terminal His-tagged fusion protein. Lane 2: C-terminal His-tagged fusion protein. The signal was developed with Goat Anti-Mouse IgG (H&L) [HRP] Polyclonal Antibody. The results show that Anti-His tag Antibody (3T670) (TMAS-00127) can recognize N-terminal and C-terminal His-tagged proteins. 9. BLI (Biolayer interferometry) binding affinity measurements of Anti-His tag Antibody (3T670) (TMAS-00127) to different His-tagged fusion proteins. The results show that Anti-His tag Antibody (3T670) (TMAS-00127) is able to recognise the His tag at different positions in the protein. |
