| Verified Activity | 1. The HepG2 (H) cells were fixed with 4% PFA (10 min at r. T.) and then permeabilized with 90% ice-cold methanol for 20 min at -20℃, the cells then were incubated in 5% BSA to block non-specific protein-protein interactions (30 min at r. T.). Primary Antibody (green): Rabbit Anti-HDAC8 antibody (TMAB-06945): 1 μg/10^6 cells; Secondary Antibody: Goat anti-Rabbit IgG-BF488: 1 μg/test. Isotype Control (orange): Rabbit IgG. Blank control (black): PBS. Acquisition of 20,000 events was performed. 2. 4% Paraformaldehyde-fixed HepG2 (H) cell; Triton X-100 at r. T. for 20 min; Antibody incubation with (HDAC8) monoclonal Antibody, unconjugated (TMAB-06945) 1: 200, 90 min at 37°C; followed by conjugated Goat Anti-Rabbit IgG antibody (green) at 37°C for 90 min, DAPI (blue) was used to stain the cell nuclei. PBS instead of the primary antibody was used as the blank control. 3. 25 μg total protein per lane of various lysates (see on figure) probed with HDAC8 monoclonal antibody, unconjugated (TMAB-06945) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r. T. for 60 min. |

| Research Background | Histones play a critical role in transcriptional regulation, cell cycle progression, and developmental events. Histone acetylation/deacetylation alters chromosome structure and affects transcription factor access to DNA. The protein encoded by this gene belongs to class I of the histone deacetylase family. It catalyzes the deacetylation of lysine residues in the histone N-terminal tails and represses transcription in large multiprotein complexes with transcriptional co-repressors. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2009]. |
| Immunogen | A synthesized peptide: human HDAC8 |
| Antigen Species | Human |
| Gene Name | HDAC8 |
| Gene ID | |
| Protein Name | Histone deacetylase 8 |
| Uniprot ID | |
| Function | Responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4). Histone deacetylation gives a tag for epigenetic repression and plays an important role in transcriptional regulation, cell cycle progression and developmental events. Histone deacetylases act via the formation of large multiprotein complexes. May play a role in smooth muscle cell contractility. |
