Verified Activity
1. Paraformaldehyde-fixed, paraffin embedded Human Lung Cancer; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with GOLPH2 Monoclonal Antibody, Unconjugated (TMAB-06636) at 1:200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L-HRP and DAB staining.
2. Paraformaldehyde-fixed, paraffin embedded Human Colon; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with GOLPH2 Monoclonal Antibody, Unconjugated (TMAB-06636) at 1:200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L-HRP and DAB staining.
3. Paraformaldehyde-fixed, paraffin embedded Human appendix; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with GOLPH2 Monoclonal Antibody, Unconjugated (TMAB-06636) at 1:200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L-HRP and DAB staining.
4. Paraformaldehyde-fixed, paraffin embedded Human Prostate; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with GOLPH2 Monoclonal Antibody, Unconjugated (TMAB-06636) at 1:200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L-HRP and DAB staining.
5. Paraformaldehyde-fixed, paraffin embedded Human Pancreatic Cancer; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with GOLPH2 Monoclonal Antibody, Unconjugated (TMAB-06636) at 1:200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L-HRP and DAB staining.
6. 25 ug total protein per lane of various lysates (see on figure) probed with GOLPH2 monoclonal antibody, unconjugated (TMAB-06636) at 1: 1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r. T. for 60 min.
7. 4% Paraformaldehyde-fixed LNCaP (H) cell; Triton X-100 at r. T. for 20 min; Antibody incubation with (GOLPH2) monoclonal Antibody, unconjugated (TMAB-06636) 1: 100, 90 min at 37°C; followed by conjugated Goat Anti-Rabbit IgG antibody (green, BF488) at 37°C for 90 min, DAPI (blue) was used to stain the cell nuclei. PBS instead of the primary antibody was used as the blank control.
8. The LNCaP (H) cells were fixed with 4% PFA (10 min at r. T.) and then permeabilized with 90% ice-cold methanol for 20 min at -20℃, the cells then were incubated in 5% BSA to block non-specific protein-protein interactions (30 min at r. T.), followed by secondary antibody incubation for 40 min at room temperature. Primary Antibody (green): Rabbit Anti-GOLPH2 antibody (TMAB-06636, 1: 100); Isotype Control (orange): Rabbit IgG. Blank control (black): PBS. Acquisition of 20,000 events was performed.

Host SpeciesRabbit
Subcellular LocalizationGolgi apparatus, cis-Golgi network membrane; Single-pass type II membrane protein. Note=Early Golgi. Cycles via the cell surface and endosomes upon lumenal pH disruption.
Tissue SpecificityWidely expressed. Highly expressed in colon, prostate, trachea and stomach. Expressed at lower level in testis, muscle, lymphoid tissues, white blood cells and spleen. Predominantly expressed by cells of the epithelial lineage. Expressed at low level in normal liver. Expression significantly increases in virus (HBV, HCV) infected liver. Expression does not increase in liver disease due to non-viral causes (alcohol-induced liver disease, autoimmune hepatitis). Increased expression in hepatocytes appears to be a general feature of advanced liver disease. In liver tissue from patients with adult giant-cell hepatitis (GCH), it is strongly expressed in hepatocytes-derived syncitial giant cells. Constitutively expressed by biliary epithelial cells but not by hepatocytes.

Research BackgroundThe Golgi complex plays a key role in the sorting and modification of proteins exported from the endoplasmic reticulum. The protein encoded by this gene is a type II Golgi transmembrane protein. It processes proteins synthesized in the rough endoplasmic reticulum and assists in the transport of protein cargo through the Golgi apparatus. The expression of this gene has been observed to be upregulated in response to viral infection.
