1. Tissue/cell: mouse heart tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer (0.01 M, pH 6.0), Boiling bathing for 15 min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30 min; Blocking buffer (normal goat serum) at 37℃ for 20 min;
Incubation: Anti-Glut12 Polyclonal Antibody, Unconjugated (TMAB-06549) 1: 200, overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining

12. 25 μg total protein per lane of various lysates (see on figure) probed with GLUT12 polyclonal antibody, unconjugated (TMAB-06549) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r. T. for 60 min.
13. The MCF-7 (H) cells were incubated in 5% BSA to block non-specific protein-protein interactions (30 min at r. T.). Primary Antibody (green): Rabbit Anti-GLUT12 antibody (TMAB-06549): 1 μg/10^6 cells; Secondary Antibody (white blue): Goat anti-Rabbit IgG-BF647: 1 μg/test. Isotype Control (orange): Rabbit IgG. Blank control (black): PBS. Acquisition of 20,000 events was performed.
