| Verified Activity | 1. 4% Paraformaldehyde-fixed Hela (H) cell; Triton X-100 at r. T. for 20 min; Antibody incubation with (Exportin 1) monoclonal Antibody, unconjugated (TMAB-05751) 1: 100, 90 min at 37°C; followed by conjugated Goat Anti-Rabbit IgG antibody (green, BF488) at 37°C for 90 min, DAPI (blue) was used to stain the cell nuclei. PBS instead of the primary antibody was used as the blank control. 2. The Hela (H) cells were fixed with 4% PFA (10 min at r. T.) and then permeabilized with 90% ice-cold methanol for 20 min at -20℃, the cells then were incubated in 5% BSA to block non-specific protein-protein interactions (30 min at r. T.). Primary Antibody (green): Rabbit Anti-Exportin 1 antibody; Secondary Antibody (white blue): Goat anti-Rabbit IgG-BF488 (BF488): 1 μg/test. Isotype Control (orange): Rabbit IgG. Blank control (black): PBS. Acquisition of 20,000 events was performed. 3. 25 ug total protein per lane of various lysates (see on figure) probed with Exportin 1 monoclonal antibody, unconjugated (TMAB-05751) at 1: 1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r. T. for 60 min. 4. Paraformaldehyde-fixed, paraffin embedded Mouse Cerebrum; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with Exportin 1 Recombinant Monoclonal Antibody, Unconjugated (TMAB-05751) at 1:200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L-HRP and DAB staining. 5. Paraformaldehyde-fixed, paraffin embedded Rat Cerebrum; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with Exportin 1 Recombinant Monoclonal Antibody, Unconjugated (TMAB-05751) at 1:200 overnight at 4°C, followed by conjugation to the Goat Anti-Rabbit IgG H&L-HRP and DAB staining. 6. | Research Background | Mediates the nuclear export of cellular proteins (cargos) bearing a leucine-rich nuclear export signal (NES) and of RNAs. In the nucleus, in association with RANBP3, binds cooperatively to the NES on its target protein and to the GTPase RAN in its active GTP-bound form (Ran-GTP). Docking of this complex to the nuclear pore complex (NPC) is mediated through binding to nucleoporins. |

| Function | Mediates the nuclear export of cellular proteins (cargos) bearing a leucine-rich nuclear export signal (NES) and of RNAs. In the nucleus, in association with RANBP3, binds cooperatively to the NES on its target protein and to the GTPase RAN in its active GTP-bound form (Ran-GTP). Docking of this complex to the nuclear pore complex (NPC) is mediated through binding to nucleoporins. Upon transit of an nuclear export complex into the cytoplasm, disassembling of the complex and hydrolysis of Ran-GTP to Ran-GDP (induced by RANBP1 and RANGAP1, respectively) cause release of the cargo from the export receptor. The directionality of nuclear export is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus. Involved in U3 snoRNA transport from Cajal bodies to nucleoli. Binds to late precursor U3 snoRNA bearing a TMG cap. Several viruses, among them HIV-1, HTLV-1 and influenza A use it to export their unspliced or incompletely spliced RNAs out of the nucleus. Interacts with, and mediates the nuclear export of HIV-1 Rev and HTLV-1 Rex proteins. Involved in HTLV-1 Rex multimerization. |
