Verified Activity | 1. PDIA4 was immunoprecipitated using: -Lane A:0.5 mg Hela Whole Cell Lysate. -Lane B:0.5 mg MCF-7 Whole Cell Lysate. -2 µL anti-PDIA4 rabbit monoclonal antibody and 60 μg of Immunomagnetic beads Protein G. -Primary antibody: -Anti-PDIA4 rabbit monoclonal antibody, at 1:100 dilution. -Secondary antibody: -Dylight 800-labeled antibody to rabbit IgG (H+L), at 1:5000 dilution. -Developed using the odyssey technique. -Performed under reducing conditions. -Predicted band size: 73 kDa. -Observed band size: 73 kDa. 2. Anti-PDIA4 rabbit monoclonal antibody at 1:500 dilution. -Lane A: A431 Whole Cell Lysate. -Lane B: NIH-3T3 Whole Cell Lysate. -Lane C: 293T Whole Cell lysate. -Lysates/proteins at 30 μg per lane. -Secondary -Goat Anti-Rabbit IgG H&L (Dylight800) at 1/10000 dilution. -Developed using the Odyssey technique. -Performed under reducing conditions. -Predicted band size:73 kDa. -Observed band size:73 kDa(We are unsure as to the identity of these extra bands.) 3. Flow cytometric analysis of Human PDIA4 expression on HepG2 cells. The cells were treated according to manufacturer’s manual (BD Pharmingen™ Cat. No. 554714), stained with purified anti-Human PDIA4, t 4. Immunofluorescence staining of PDIA4 in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti-Human PDIA4 monoclonal antibody (dilution ratio 1:60) at 4°C overnight. Then cells were stained with the Alexa Fluor®488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI (blue).Positive staining was localized to Cytoplasm. |

Research Background | ERP72, also known as PDIA4, is an endoplasmic reticulum luminal protein which belongs to the protein disulfide isomerase family. ERP72 is a stress protein and participates in the catalysis of protein-S-S-bond rearrangement. Both PDIA4 and PDIA3 function as proteases, protein disulfide isomerases, phospholipases or an arrangement of these. ERP72 compose part of a large chaperone multiprotein complex comprising CABP1, DNAJB11, HSP90B1, HSPA5, HYOU, PDIA2, PDIA4, PPIB, SDF2L1, UGT1A1 and very small amounts of ERP29, but not, or at very low levels, CALR nor CANX. |
