Mayserve as a rigid scaffold in the complex and may contribute tocatalysis through positioning of the substrate and theubiquitin-conjugating enzyme. The E3 ubiquitin-protein ligaseactivity of the complex is dependent on the neddylation of thecullin subunit and is inhibited by the association of thedeneddylated cullin subunit with TIP120A/CAND1 (By similarity). Thefunctional specificity of the ECS complex depends on the substraterecognition component. ECS(VHL) mediates the ubiquitination ofhypoxia-inducible factor (HIF).
