Paraformaldehyde-fixed, paraffin embedded (human lung carcinoma); Antigen retrieval by boiling in sodium EDTA buffer (Ph9.0) for 15 min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30 min; Incubation with (Collagen IV) Monoclonal Antibody, Unconjugated (TMAB-04525) at 1:500 overnight at 4°C, followed by operating according to SP Kit (Rabbit) instructions and DAB staining.

Type IV collagen is the major structural component of glomerular basement membranes (GBM), forming a 'chicken-wire' meshwork together with laminins, proteoglycans and entactin/nidogen.
