| Verified Activity | 1. Paraformaldehyde-fixed, paraffin embedded (human neurilemmoma); Antigen retrieval by boiling in sodium citrate buffer (pH6) for 15 min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30 min; Antibody incubation with (CLIC6) Polyclonal Antibody, Unconjugated (TMAB-04453) at 1: 200 overnight at 4°C, followed by a conjugated secondary at [1:500] for 90 minutes and DAPI staining of the nuclei. 2. Blank control: Mouse kidney. Primary Antibody (green line): Rabbit Anti-CLIC6 antibody (TMAB-04453) Dilution: 2 μg /10^6 cells; Isotype Control Antibody (orange line): Rabbit IgG. Secondary Antibody: Goat anti-rabbit IgG-AF488 Dilution: 1 μg /test. Protocol The cells were fixed with 4% PFA (10 min at room temperature) and then permeabilized with 0.1% PBST for 20 min at room temperature. The cells were then incubated in 5% BSA to block non-specific protein-protein interactions for 30 min at room temperature. Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed. 3. Blank control: Mouse kidney. | Research Background | CLIC6 (chloride intracellular channel 6) is believed to play a critical role in water-secreting cells, possibly through the regulation of chloride ion transport. The CLIC6 gene is a rare example of large-scale segmental paralogy in which a large (approximately 500 kb) segment on human chromosome (HC) 21 (21q22) is triplicated on HC 1 and HC 6. CLIC6 is also known to interact with dopamine receptors DRD2, DRD3 and DRD4. CLIC6 is primarily expressed in the cytoplasm, however, upon chloride ion efflux from the cell, CLIC6 is translocated to the plasma membrane. CLIC6 has been identified in brain, placenta, pancreas and liver. |
