| Research Background | The regulation of the intracellular concentration of calcium is important for proper maintenance of voltage-gated ion channels which control muscle and nerve function. Calcium homeostasis is regulated by a variety of proteins. CHERP (calcium homeostasis endoplasmic reticulum protein), also known as SRA1, DAN16 or SCAF6, is a 916 amino acid protein that localizes to the cytoplasm and the endoplasmic reticulum (ER). Expressed in pancreas, brain, lung, placenta, liver, kidney, heart and skeletal muscle, CHERP is involved in maintaining calcium homeostasis and plays a role in cell growth and proliferation. CHERP contains one G-patch domain, one RPR domain and one SURP motif and is expressed as two isoforms due to alternative splicing events. |

| Function | The Calcium homoeostasis endoplasmic reticulum protein (CHERP) gene has been recently identified and localized on chromosome 19p13.1. CHERP has high homology to an SR related CTD protein, which is known to interact with the largest subunit of RNA polymerase II. CHERP is an integral endoplasmic reticulum membrane protein is involved in calcium mobilization induced by thrombin. Confocal microscopy revealed that CHERP is associated with Ins(1,4,5)P3 receptor throughout the cytoplasm and perinuclear region in Jurkat T lymphocyte. CHERP Anti sense treatment induced a decrease in CHERP, which caused an impaired increase in free cytoplasmic calcium but calcium influx remains unaffected, with some deficient in endoplasmic reticulum calcium stores. In the CHERP depleted Jurkat T lymphocytes the calcium dependent translocation of nuclear factor of activated T cells (NFAT) from cytoplasm to nucleus was also suppressed with significant suppression of cell. |
