| Verified Activity | 1. Immunofluorescence staining of Hela cell with TMAH-00260 at 1:150, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was FITC-conjugated AffiniPure Goat Anti-human IgG(H+L). 2. Immunofluorescence staining of MCF7 cell with TMAH-00260 at 1:150, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was FITC-conjugated AffiniPure Goat Anti-human IgG(H+L). 3. Overlay Peak curve showing Hela cells surface stained with TMAH-00260 (red line) at 1:100. Then 10% normal goat serum was Incubated to block non-specific protein-protein interactions followed by the antibody (1µg/1*10^6 cells) for 45 min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-human IgG(H+L) at 1/200 dilution for 35 min at 4°C. Isotype control antibody (green line) was human IgG1 (1µg/1*10^6 cells) used under the same conditions. Acquisition of >10,026 events was performed. |

| Research Background | Cell surface glycoprotein that plays a role in cell adhesion, intracellular signaling and tumor progression. Mediates homophilic and heterophilic cell adhesion with other carcinoembryonic antigen-related cell adhesion molecules, such as CEACAM6. Plays a role as an oncogene by promoting tumor progression; induces resistance to anoikis of colorectal carcinoma cells. (Microbial infection) Receptor for E.coli Dr adhesins. Binding of E.coli Dr adhesins leads to dissociation of the homodimer. |
