| Verified Activity | Immunofluorescence staining of TBXT in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with mouse anti-Human T monoclonal antibody (dilution ratio 1:60) at 4°C overnight. Then cells were stained with the Alexa Fluor®488-conjugated Goat Anti-mouse IgG secondary antibody (green). Positive staining was localized to Nucleus. |

| Research Background | Brachyury is encoded by the TBXT gene. TBXT plays in the Cnidaria, appears to be in defining the blastopore. Brachyury is an important factor in promoting the epithelial-mesenchymal transition (EMT). Cells that over-express brachyury have down-regulated expression of the adhesion molecule E-cadherin, which allows them to undergo EMT. This process is at least partially mediated by the transcription factors AKT and Snail. Brachyury is overexpressed in a number of tumor types. |
