1. RAW264.7 (M) cells were treated with or without LPS (1 μg/ml) for 2 h, 25 μg total protein per Lane of cell lysates probed with ATF3 polyclonal antibody, unconjugated (TMAB-00158) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at RT for 60 min.
2. Paraformaldehyde-fixed, paraffin embedded Human Placenta; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; The section was incubated with ATF3 Polyclonal Antibody, Unconjugated (TMAB-00158) at 1:200 overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining.

This protein binds the cAMP response element (CRE) (consensus: 5'-GTGACGT[AC][AG]-3'), a sequence present in many viral and cellular promoters. Represses transcription from promoters with ATF sites. It may repress transcription by stabilizing the binding of inhibitory cofactors at the promoter. Isoform 2 activates transcription presumably by sequestering inhibitory cofactors away from the promoters.
