| Verified Activity | 1. Blank control: Mouse splenocytes (blue) Isotype Control Antibody: Rabbit IgG (orange); Secondary Antibody: Goat anti-rabbit IgG-FITC (white blue), Dilution: 1:100 in 1 X PBS containing 0.5% BSA; Primary Antibody Dilution: 1 μl in 100 μL 1X PBS containing 0.5% BSA (green). 2. Blank control: A431. Primary Antibody (green line): Rabbit Anti-ADAM8 antibody (TMAB-02294) Dilution: 2 μg /10^6 cells; Isotype Control Antibody (orange line): Rabbit IgG. Secondary Antibody: Goat anti-rabbit IgG-AF488 Dilution: 1 μg /test. Protocol The cells were incubated in 5% BSA to block non-specific protein-protein interactions for 30 min at at room temperature. Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed. 3. Black line : Positive blank control A431); Negative blank control (HepG2) Green line : Primary Antibody (Rabbit Anti-ADAM8 antibody (TMAB-02294)) Orange line:Isotype Control Antibody (Rabbit IgG). Blue line : Secondary Antibody (Goat anti-rabbit IgG-AF488) A431 (Positive) and HepG2 (Negative control) cells (black) were incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with ADAM8 Antibody (TMAB-02294) at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2% BSA in PBS, followed by secondary antibody (blue) incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange). 4. 4% Paraformaldehyde-fixed Raji (H) cell; Antibody incubation with (ADAM8) polyclonal Antibody, unconjugated (TMAB-02294) 1:100, 90 min at 37°C; followed by conjugated Goat Anti-Rabbit IgG antibody (green) at 37°C for 90 min, DAPI (blue) was used to stain the cell nuclei. PBS instead of the primary antibody was used as the blank control. |

The extracellular region of ADAM8 shows significant amino acid sequence homology to hemorrhagic snake venom proteins, including the metalloprotease and disintegrin domains. The expression of ADAM8 is upregulated by retinoic acid and vitamin D3. |
