1. Paraformaldehyde-fixed, paraffin embedded (Human cervical cancer); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30 min; Antibody incubation with (Actin, alpha skeletal muscle) Monoclonal Antibody, Unconjugated (TMAB-02258) at 1:400 overnight at 4°C, followed by a conjugated secondary for 20 minutes and DAB staining.
2. Paraformaldehyde-fixed, paraffin embedded (rat skeletal muscle); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30 min; Antibody incubation with (Actin, alpha skeletal muscle) Monoclonal Antibody, Unconjugated (TMAB-02258) at 1: 200 overnight at 4°C, followed by operating according to SP Kit (Mouse) instructions and DAB staining.

Research BackgroundThe product encoded by this gene belongs to the actin family of proteins, which are highly conserved proteins that play a role in cell motility, structure and integrity. Alpha, beta and gamma actin isoforms have been identified, with alpha actins being a major constituent of the contractile apparatus, while beta and gamma actins are involved in the regulation of cell motility. This actin is an alpha actin that is found in skeletal muscle. Mutations in this gene cause nemaline myopathy type 3, congenital myopathy with excess of thin myofilaments, congenital myopathy with cores, and congenital myopathy with fiber-type disproportion, diseases that lead to muscle fiber defects.
