Targeted covalent inhibitors (TCIs) have the ability to enhance the potency and/or selectivity of small-molecule inhibitors by incorporating reactive functional groups designed to covalently bind to specific sites within a target. These inhibitors contain specific functional moieties engineered to react with corresponding sites on the target, typically amino acid side chains. These side chains can either be part of the target enzyme's catalytic mechanism or non-catalytic residues located near the binding pocket. Since covalent bonds are significantly stronger than non-covalent interactions, the development of covalent inhibitors offers the potential for greater potency compared to their non-covalent counterparts. These covalent bonds are typically irreversible; therefore, covalent inhibitors provide a longer duration of action than reversible inhibitors, as the target remains inhibited until the protein is degraded and subsequently resynthesized.
To date, most research has focused on targeting non-catalytic cysteine residues, such as those found in cysteine proteases and protein kinases, where achieving selectivity for specific members of a protein family can be challenging. Cysteine is characterized by its relatively low abundance and high nucleophilicity among proteins. The thiol form of cysteine has been shown to form covalent bonds with various electrophilic warheads, exhibiting broad reactivity, such as forming adducts with Michael acceptors.
