Description
(R)-Chloro-butylphthalide-O-NBP is a chiral derivative of 3-butylphthalimide and serves as a neuroprotective agent. It exhibits strong blood-brain barrier (BBB) protective properties. In vivo experiments demonstrate that (R)-Chloro-butylphthalide-O-NBP significantly reduces infarct volume and effectively preserves the integrity of the BBB. This compound is applicable in ischemic stroke research.
In vitro
(R)-Chloro-butylphthalide-O-NBP (compound R-30) at concentrations of 0.1 μM, 1 μM, and 10 μM significantly enhances the survival rate of PC12 cells under OGD/R conditions. Additionally, the same concentrations of (R)-Chloro-butylphthalide-O-NBP markedly reduce LDH release in PC12 cells subjected to hypoxia/reoxygenation injury.
In vivo
The compound (R)-Chloro-butylphthalide-O-NBP (compound R-30), when administered intravenously at doses of 5-20 mg/kg, has been shown to decrease infarct volume, improve neurological deficit scores, and preserve blood-brain barrier integrity in a rat model of MCAO ischemic stroke. Following a single intravenous administration of (R)-Chloro-butylphthalide-O-NBP at doses of 100-200 mg/kg, rats showed no significant abnormalities in body weight, food consumption, post-mortem organ weight, or biochemical parameters.
