| In vitro | Four isoforms of Pyruvate kinase (PK) are found in mammals: L and R isoforms, produced by the PKLR gene, are expressed in the liver and red blood cells, respectively. The PKM gene is responsible for two splice variants: M1, prevalent in most adult tissues, and M2, found during embryonic development and in specific adult tissues, such as the kidney and hematopoietic stem cells. Additionally, many tumor cells express PKM2. This tetrameric isoform, which is allosterically regulated, is naturally predisposed to suppress its activity through several mechanisms: post-translational modifications, allosteric alterations by small molecules including certain amino acids, and conversion into a dimeric form. These processes lead to a partial blockade of glycolysis at its final stage[1]. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
