Fomivirsen sodium (ISIS-2922 sodium) is a 21-base phosphorothioate oligodeoxynucleotide that is complementary to the mRNA of the major immediate early region (IE2) of human cytomegalovirus (CMV). It inhibits viral replication by cleaving the target mRNA through the ribonuclease H (RNase H) mechanism. Description
Fomivirsen sodium (ISIS-2922 sodium) is a 21-base phosphorothioate oligodeoxynucleotide that is complementary to the mRNA of the major immediate early region (IE2) of human cytomegalovirus (CMV). It inhibits viral replication by cleaving the target mRNA through the ribonuclease H (RNase H) mechanism. Following intravitreal administration, it can be used in research related to cytomegalovirus retinitis associated with AIDS.
Targets & IC50
MRC5 cells:0.2 μM, RPE cells:0.03 μM
In vitro
Method: Human retinal pigment epithelial (RPE) cells and human fibroblast cells (MRC-5) were treated with fomivirsen. Inhibition of CMV replication was measured, and the IC50 was calculated.
Result: Fomivirsen dose-dependently inhibited CMV replication in both RPE cells and MRC-5 cells. The mean IC50 was 0.03 μM in RPE cells and 0.2 μM in MRC-5 cells [1].
In vivo
Method: A single intravitreal injection of 66 μg fomivirsen was administered into rabbit eyes. Vitreous clearance was measured, and the half-life was calculated following first-order kinetics.
Result: The clearance half-life of fomivirsen from the vitreous in rabbit eyes was 62 hours. At day 10, 22% of the total radioactivity remained in the vitreous [1].

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
