Description
FLT3-ITD-IN-3 (13v) is an orally active inhibitor of FLT3-ITD (internal tandem duplication of FLT3), which works by blocking FLT3 signaling. This inhibition leads to cell cycle arrest in the G0/G1 phase and induces apoptosis. The compound is currently employed in studies related to acute myeloid leukemia (AML).
In vitro
FLT3-ITD-IN-3 (13v) is a potent and selective inhibitor that suppresses the proliferation of three cell lines with the FLT3-ITD mutation (MOLM-14, MOLM-13, and MV4-11) with IC50 values of 0.59 μM, 0.14 μM, and 0.41 μM, respectively. Additionally, FLT3-ITD-IN-3 (13v) significantly reduces the phosphorylation of FLT3 and STAT5 in a dose-dependent manner.
In vivo
FLT3-ITD-IN-3 (13v) shows optimal peak plasma concentration (C max = 306.1 ng/mL) and has a moderate half-life (T 1/2 = 1.83 h).
