| In vitro | I. Preparation of stock solution Dissolve 1 mg BODIPY 581/591 C11 in 0.1983 mL DMSO to obtain 10 mM BODIPY 581/591 C11. Note: Store the stock solution at -20°C or -80°C away from light and avoid repeated freezing and thawing. Preparation of BODIPY 581/591 C11 working solution Dilute the stock solution with pure DMEM medium or PBS to make BODIPY 581/591 C11 working solution at 1-10 μM. Note: Please adjust the concentration of BODIPY 581/591 C11 Working Solution according to your experimental plan and practice. Cell Staining 1) Preparation requirements for different cell types: Suspension cells: centrifuge at 4℃, 1000g for 3-5 minutes, discard the supernatant. wash with PBS twice for 5 minutes each time. Adherent cells: discard the cell culture medium, add trypsin (1 ½ min) to dissociate the cells and make a single cell suspension. Centrifuge at 1000 g for 3-5 minutes at 4°C, discard supernatant and wash twice with PBS for 5 minutes each time. Note: Please be gentle when using PBS cells and when blowing the cells with the pipette gun. 2) Add 1 mL of BODIPY 581/591 C11 working solution to the prepared cells and incubate for 30 minutes at room temperature. 3) Centrifuge at 400 g for 4 minutes at 4°C and discard the supernatant. 4) Wash with PBS 2 times for 5 minutes each time. 5) Resuspend the cells using pure DMEM medium or PBS, and observe the stained cells by fluorescence microscope or flow cytometer. Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
