| In vitro | Methods: Seven AML cell lines and primary CD34+CD38- leukemic stem cells (LSC) were treated with AOH1996 at concentrations of 0.5–1 μM for 24 hours, and cell proliferation, apoptosis, and colony formation assays were performed. Results: AOH1996 dose-dependently inhibited proliferation and colony formation of AML cells and LSC, induced apoptosis, and had no significant effect on normal CD34+CD38- hematopoietic stem cells.[1] |
| In vivo | Methods: Human primary AML cells were transplanted into Es1(ko)SCID mice, and 7 days later, AOH1996 (100 mg/kg) was administered by oral gavage twice daily for 3 weeks. Results: AOH1996 significantly reduced leukemia burden in peripheral blood and spleen, and extended median survival from 35 days to 50 days; secondary transplantation confirmed reduced LSC burden, with survival extended from 30 days to 42 days.[1] |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
