Description
Anti-MRSA agent 11 demonstrates strong activity against both fluoroquinolone-sensitive strain USA500 and resistant MRSA strain Mu50, with a MIC value of 0.39 μg/mL. It also possesses a favorable in vivo half-life and safety profile.
In vitro
Anti-MRSA agent 11, at a concentration of 100 μM for 24 hours, shows no cytotoxic effects on A549 and HEK-293 cells, demonstrating high selective toxicity towards bacterial cells. Additionally, at 0.25 μg/ml over 15 generations, it maintains sustained antibacterial activity against the MRSA strain ATCC33591. This compound effectively penetrates bacterial cells through their membranes, particularly the outer membrane of Gram-negative bacteria, to exert its function. Anti-MRSA agent 11 inhibits bacterial topoisomerase IV, thereby disrupting the process of genetic material transmission in bacteria.
In vivo
Compound 11, used as an anti-MRSA agent at doses of 0, 400, 1000, and 2000 mg/kg by oral administration for 8 consecutive days, did not significantly affect the body weight or food intake of mice.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
