| Verified Activity | 1. 4% Paraformaldehyde-fixed Hel.92.1.7 (H) cell; Triton X-100 at r. T. for 20 min; Antibody incubation with (GATA1) monoclonal Antibody, unconjugated (TMAB-06350) 1:100, 90 min at 37°C; followed by conjugated Goat Anti-Rabbit IgG antibody (green) at 37°C for 90 min, DAPI (blue) was used to stain the cell nuclei. PBS instead of the primary antibody was used as the blank control. 2. 25 μg total protein per lane of various lysates (see on figure) probed with GATA1 monoclonal antibody, unconjugated (TMAB-06350) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r. T. for 60 min. |

| Research Background | GATA1 (Globin transcription factor 1) is a Cys2/Cys2 zinc finger DNA binding protein that is expressed primarily in erythroid, megakaryocytic, mast cells and eosinophilic cells. It belongs to the GATA family of transcription factors. GATA1 is a transcriptional activator which probably serves as a general switch factor for erythroid development. It binds to DNA sites with the consensus sequence [AT]GATA[AG] within regulatory regions of globin genes and of other genes expressed in erythroid cells. The protein also plays an important role in erythroid development by regulating the switch from fetal hemoglobin production to adult hemoglobin. |
